Clutching at Guidance Cues: The Integrin–FAK Axis Steers Axon Outgrowth

نویسندگان

چکیده

Integrin receptors are essential contributors to neurite outgrowth and axon elongation. Activated integrins engage components of the extracellular matrix, enabling growth cone form point contacts, which connect substrate dynamic intracellular protein complexes. These adhesion complexes facilitate efficient migration extension. Major signalling pathways mediated by complex instigated focal kinase (FAK), whilst axonal guidance molecules present in vivo promote turning or retraction local modulation FAK activity. Activation is marked phosphorylation following integrin engagement, this activity tightly regulated during outgrowth. inhibition slows reducing contact turnover; however, mutant constructs with enhanced stimulate aberrant Importantly, a major structural component maturing sites, provide platform for actin polymerisation drive leading edge advance. In review, we discuss coordinated FAK, as well their role regulating We also importance integrin–FAK axis vivo, expression activation key determinants successful regeneration injury.

Translating Axon Guidance Cues

Recent studies suggest that local translation of mRNA within axons plays a key role in two aspects of growth cone navigation. One line of evidence implies a local synthesis of proteins that mediate growth cone collapse and turning in response to guidance cues; another indicates that growth cones possess the machinery that would allow the local synthesis of receptors for axon guidance cues.

Axon guidance cues in auditory development.

The innervation of the cochlear sensory epithelium is intricately organized, allowing the tonotopy established by the auditory hair cells to be maintained along the ascending auditory pathways. These auditory projections are patterned by several gene families that regulate neurite attraction and repulsion, known as axon guidance cues. In this review, the roles of various axon guidance molecules...

Morphogens as conserved axon guidance cues.

Morphogen family proteins are now widely appreciated as axon guidance cues. Because their roles as morphogens are highly conserved across phylogeny, their functional conservation in axon guidance is now being rigorously examined. Recent studies suggest that morphogens are important in shaping topographic projections in chick and Drosophila visual systems, a process that occurs even later in dev...

The growth cone cytoskeleton in axon outgrowth and guidance.

Axon outgrowth and guidance to the proper target requires the coordination of filamentous (F)-actin and microtubules (MTs), the dynamic cytoskeletal polymers that promote shape change and locomotion. Over the past two decades, our knowledge of the many guidance cues, receptors, and downstream signaling cascades involved in neuronal outgrowth and guidance has increased dramatically. Less is know...

PHRs: bridging axon guidance, outgrowth and synapse development.

Axon guidance, outgrowth, and synapse formation are interrelated developmental events during the maturation of the nervous system. Establishing proper synaptic connectivity requires precise axon navigation and a coordinated switch between axon outgrowth and synaptogenesis. The PHR (human Pam, mouse Phr1, zebrafish Esrom, DrosophilaHighwire, and C. elegansRPM-1) protein family regulates both axo...
